WHY METFORMIN

Metformin may target pathways linked to biological aging.

GPD1 Pathway

Researchers found that genetic mimicry of metformin's GPD1 target was associated with a biological age equivalent approximately 5.3 years younger.

PhenoAge

In a study of over 300,000 people, metformin users had a biological age 0.36 years younger than non-users — measured by PhenoAge, a blood-based marker of how fast the body is aging internally.

AMPKγ2

Another metformin-related pathway, AMPKγ2, was associated with a biological age equivalent approximately 4.9 years younger.

HOW METFORMIN WORKS

Activates AMPK.

Mimics fasting. Metformin works on an energy-sensing pathway in your cells. This can support: blood sugar control, cellular aging, and metabolic health.

Metformin

Activates AMPK

Metformin activates AMPK, a cellular switch that turns on when energy runs low. When it's on, the body shifts into repair mode, using energy more efficiently and reducing excess glucose production.

Reduces glucose

Metformin can reduce the liver's tendency to release sugar into the bloodstream between meals. The result is steadier blood sugar throughout the day and less strain on insulin production.

Suppresses mTOR

AMPK activation signals the body to dial back mTOR, the pathway that drives cell growth. Keeping mTOR in check is associated with slower cellular aging, and mirrors what the body does during caloric restriction.

What is metformin?

Metformin is an FDA-approved medication for type 2 diabetes. Off-label, doctors sometimes prescribe it within a longevity framework given growing evidence of its effects on the cellular pathways associated with biological aging.

How do you take metformin?

Metformin is an oral tablet taken with meals. Always follow the dosage instructions from your doctor.

What are the side effects of metformin?

Common side effects include nausea, loose stools, and stomach discomfort, especially in the first few weeks. These should settle as your body adjusts. Contact your doctor if side effects are persistent or bothersome.

Who shouldn't take metformin?

Metformin is not recommended for people with severe kidney impairment, a known hypersensitivity to metformin, or acute metabolic acidosis including diabetic ketoacidosis. Extra caution applies to patients over 65 and those scheduled for imaging procedures using iodinated contrast. Your doctor will review your kidney function and full health history before prescribing.
